The post title specifies 2016. This is the design in production since the 2011 model year. Earlier Explorers share nothing but the name, so experience with them will not be relevant

That sounds like a workable combination, and the towing capacity doesn't change with 2WD or 4WD.
Either of the V6 engines leads to essentially the same towing capacity, but the 3.5 EcoBoost only comes in 4WD. The EcoBoost also has more power (which helps performance, but not towing capacity), and is turbocharged so a given level of power is available at a lower speed than the non-turbo engine.
If you are interested in a lower vehicle on the same platform, also available with the 3.5 EcoBoost, you might consider the Ford Flex... which has the same V6 engines and may have the same towing capacity (the online specs say "up to Class III")

The 2.7 L EcoBoost is not offered in the Explorer - that model comes with the 3.5L EcoBoost V6, the 2.3L EcoBoost 4-cylinder, and a 3.5L non-turbo V6. The 2.7 EcoBoost is a V6, closely related to the 3.5 EcoBoost and unrelated to the 2.3 EcoBoost.
Even the 2.3 EcoBoost has enough power, but power is usually not the limitation to towing capacity. This limitation with this engine may be the transmission or cooling system.
