A hose over a hose barb, held with a clamp, should not have any sealant. The hose may still be difficult to remove, since it will be formed to match the barbs by continued clamping.
My only hint for this hose removal is to push the end of the hose off, rather than just pulling on the hose (which tightens the hose onto the barb). Hose grip pliers can be helpful, too. Breaking the barbed fitting would be undesirable, particularly if it is directly bonded into the tank (rather than being screwed into a threaded bung)... that's the thing that always makes me hesitate to mess with this sort of connection.
